PERSIAN Biobank

Developing a biobank— a type of repository storing biological samples—has become an important part of clinical research in the past few decades. If stored under the appropriate conditions, a variety of biological specimens can be kept for many years, and be used for research in medical, epidemiological, and genetic studies, ideally to identify new biomarker, onco-markers or genes causing disease in a large section of the population.

A variety of samples are stored in the PERSIAN Cohort biobank. Samples are taken at enrollment. If one of the study’s main outcomes of interest is noted in the follow up phase (cancer or other disease development), additional blood samples are taken and stored. A fraction of aliquoted samples are stored in local biobanks at the cohort centers, and a fraction is stored centrally in the PERSIAN Cohort Central Biobank. The PRESIAN Cohort Laboratory Information Management System is designed and used for easy tracking and access to samples.

Types of samples collected and stored in the PERSIAN Cohort Biobanks

Blood

Twenty milliliters (20 cc) of blood is taken from each participant (fasted). A portion of the serum is used to run the following biochemistry tests:

  • Complete blood count
  • Fasting blood sugar
  • Total cholesterol
  • HDL cholesterol
  • Triglycerides
  • Alanine transaminase
  • Aspartate transaminase
  • Alkaline phosphatase
  • Gamma-glutamyl transpeptidase
  • Blood urea nitrogen
  • Creatinine

Results obtained from the tests above are stored in the PERSIAN Cohort databases and also given back to participants as an incentive.

The majority of the blood taken is aliquoted and stored in -70C freezers. Blood is stored as whole blood, red blood cells, plasma, serum and buffy coat.

Urine

Urine is collected from each individual and stored in -30C freezers. A urine analysis is also performed, the results of which are recorded in the cohort database and also returned to participants.

Hair and Nail

All finger and toenails as well as 200-300 strands of hair are collected from each participant. Samples are placed inside foil wraps, which are then placed in individual zipped bags along with a desiccant de-humidifier, and kept in cool, dry storage.

Other Samples

In addition to the above samples, the PERSIAN Birth Cohort collects additional samples such as cord blood, maternal milk, stool, and baby’s first tooth from a subset of participants.
